You can't take the door off. Not without disassembling the camera.
Some system cameras has a small spring-supported pin that holds the door. This is to make room for stuff like film magazines as shown here:

Turn the setting in the drive mode ( Top cover LCD Panel ) from C to S This mode with only let you take one shot at a time as you lift you finger up and down on the release button.

The Nikon N6006 will use just about any Nikon F-mount lens made in the past 55 years. About the only exceptions are some rare (and very expensive) fisheye lenses and the newer DX lenses specifically designed for digital cameras.

Yes, the autofocus should work. However you'll likely get vignetting on your photos because a DX lens is designed to cover a smaller area than a frame of 35mm film.

The N6006 uses one DL223A or CR-P2 battery.

If you have more then one autofocus lens, is the problem limited to one lens only? if it turns out that the camera needs professional repair (and it may), keep in mind that an N6006 body is currently available on E-bay for $35 (buy it now price) and an N8008 is only $40 (buy it now).

What speed film? Does the roll have the DX markings (the black-and-silver rectangles)? If it doesn't, what happens if you set the film speed manually?

There is a safety switch to prevent accidental operation.  Hold the camera like you are taking a photo.  Rotate the camera down so the lense is pointing towards the floor and you are now looking at the bottom of the camera.  On the right is a small button.  In the middle is a slide switch with an arrow pointing to the left.  You need to hold in the button on the right while sliding the switch to the left.
