We made agar gel and put it in 30 Petri dishes, but somehow they all got contaminated. Do you know what would have caused this?

We laid the dishes on the counter before we poured the agar in, and we used a glass measurer to pour it in, and now my project failed because there is stuff growing in all 30 of them.

They may be contaminated during sterilization by using autoclave

Either the water filter isn't seated properly or there is a crack in the tubing or the water line isn't seated properly in the coupling behind the grill at the freezer door. Also, inspect for breakage in the line in this area as well (coupling area).

I had the same problem. How Frustrating. What I discovered is if you pour the water in really slow, the bubble do not appear.

This was one of 3 problems I had this this maker. The other two were.

  • Could never get the clock set correctly. The buttons are very cheaply made. Would push the time advance button and the numbers would not change. Started acting whacky, flipping between the hours, minutes and program functions.
  • For 3 mornings straight the removal of pot lid gave me troubles. Was like trying to crack a safe. Needless to say, after the forth morning attempt, it lost the battle and broke.
I have been a Krups fan for many many years, dating back to the early 80s. After this even, I have become VERY disappointed with the quality of their consumer product line.

Ether the washer is over filling or has warn out the seals. If it has been abused (overloaded) it needs serious work. other wise the sensor may be stuck. Try 1/2 cup of vinegar with a load and see if that cleans the sensor travel area past the drum but inside the tub. This is a common problem for an old machine. Best leak is on the door and you may be able to tighten the screws but be careful it could mis aligne it so tighten a small amount and test by closeing the door. Do this at least twice before turning the screws hard.

How mushroom spawn is made

Make your spawn. Place small pieces of mushrooms on an agar medium in petri dishes, keep it moistened and under the proper temperatures for those particular mushrooms; the mycelium (threadlike parts that deliver nutrients) will grow out and fill the dish in about two weeks. Agar medium is a gelatinous substance made from seaweed cells that is commonly used in laboratory work.
  • Transfer the mycelium when it has just filled the petri with long, strands. Don't let the mycelium grow too long in the petri dishes. Transfer the mycelium to the sterilized grain in jars. Allow it to colonize for about two to four weeks under the proper temperature for the particular species of mushroom
  • Observe your jars. When they are filled with healthy, white filaments, you have achieved grain spawn. Mushrooms will grow right out of the jar of grain spawn, or you can use it to inoculate more grain. If you repeat the process, one jar of grain spawn can produce up to ten more.

    Edu science 600x microscope: no instruction manual and I cannot see anything through the lens (only a blurred blue light). What must I do to be able to see something ? Thx

    Well I have the instruction manual, but it is not too helpful. When I was a child I put fleas and ants on the slides, but I guess this microscope is different. Here is what my manual says.
    How to make a prepared slide: If the given sample is not thin and transparent, it cannot be observed by the microscope as the light from the reflector or the light source does not pass through it. Fibers, pollen, wool, or salt can be observed easily and a cover glass is optional. Clear samples are stained first with a drop or 2 of methylene blue, Eosin or other dying solutions (not included).
    1. TEMPORARY MOUNT: Wipe the slide and cover glass clean. Slice a thin sample with the micro-slicer (blade is very sharp...adult supervision). Pick up the sample with tweezers and put it on the center part of the glass slide. Put one drop of water on the sample using the needle that came with your kit.
    2.PERMANENT MOUNT:Proceed as above, however, before covering the slide with the cover glass, add a few drops of gum media (not included, available at the store) or Canada balsam solution or transparent adhesive glue with a disecting needle to the slide. Push down on the cover glass with the tweezers to fix it in place and leave to dry for a day.
    VIEWING LIVE BRINE SHRIMP:
    1.Pour a small amout of brine shrimp eggs into the petri dish.
    2.Add enough water so that the eggs are submerged and add just a dash of table salt.
    3.Place the petri dish in a well lit area (under a lamp with a 60 watt bulb is good, for 24 hours).
    4. When you see movement in the petri dish place it on your microscope stage, turn on the light and watch the brine shrimp in action.

    Even after a wash-a white detergent residue is left.

    I have a Whirlpool Gold dishwasher and had the same trouble. I found the solution. Fill the detergent dispenser to the top with Cascade Complete liquid gel - it is in a green bottle, then put a half a cup of white vinegar in the bottom of the tub before you start the dishwasher. I have tried them all and Cascade Complete Gel works best. Don't use powder. It really builds-up on my dishes and only the gel keeps the film off my dishes.

    I also add Jet Dry in the rinse-aid dispenser but I have a friend who use vinegar in that too. The dishes come out sparkling clean with no film or white build-up. If they still are filmy after three washes using this method, then you may have some build-up in the dishwasher and you can put a cup or two of white vinegar in the bottom - with no detergent or dishes and wash and rinse well. That helps get rid of build-up.

    Fuel becomes like a gel substance if the bike was not used for some time. There may be some gel blocking the main jet of your carburetor. If the gel formed in the tank, it may take a while to reach the carburetor. That may be why it operated without a problem for a while.

    Dishwasher

    Make sure not to prewash your dishes before you put them in the machine. The water needs to be dirty to activate the enzymes in the detergent. Are the dishes coming out cloudy? Also a sign of prewashing your dishes. Use only powder detergent and clean your machine by running 2 cups of vinegar on hottest cycle once a month. No gel and no tabs. They cause a build up in the unit and causes poor performance. Use rinse aid. Hope this helps.

    I have a residue on my dishes after washing.

    It really helps to know the Make and Model of your dishwasher.
    Recommend either Electrosol or Cascade powder. Do not use gel or tablets in any dishwasher.
    If problems continue, have the heating element checked because that is another common reason for unclean dishes.

    Dish washer will open and let the gel pack out.

    The gel packs are not supposed to go inside the dispensor. It is for powders & liquids. The gel packs & blocks just go into the dishwasher, per directions on package. They either go into the bottom or with the silverware.
