The only way it will run backwards is the timing is incorrect. If you had the flywheel or anything in the engine apart relating to the ignition pickup coils, they didnt get put back in correctly.
It's possible something is loose, and sheared the flywheel key. 2 cycle will run regardless of direction due to no valves.

First, see what you dont have.
You need good compression
Does it have a good fat blue spark at the plug.
Are you getting fuel to the spark plug.
Any bike that sits for half a year could have bad fuel, and the carburator should be cleaned, as it might be gummed up. Changing the fuel in the tank will also help.

Most likely a combination of sparkplug heat range (type of plug) and jetting, needle or air mixture settings in the carburetor. Assuming good condition of air filter and operation of the choke plunger. Are you using the correct octane fuel and fuel/oil ratio? Give the plug a clean and emery the spark contacts, set gap at 0.7mm., ideally take the plug to a Yammy dealer and check for correct range for your bike in the area where you ride.
Best to have a range of spare clean plugs handy for wet or hot days etc.
The jetting maybe dirty or wrong size too. In the meantime move the throttle needle clip down one notch. Refer to the site below for a diagram of the carburetor parts. Cheers.

SOURCE: Pw50 starts first time everytime but will not
PW have the stop\start\run switch and will not rev in the start position, there may be a problem in this switch, wire, or the ignition box which controlls this.
The small 50s can also build up carbon in the exhaust port, or the exit end of the muffler.
They also need the original air filter and cover (dont know why, but they wont rev without it).
I realize you may have checked some of these, but not much else goes wrong
